Studies on Pharmacological Screening Method of Experimental Allergic Encephalomyelitis by GPC phosphocholine phosphodiesterase Activity in Mice


Abstract
Experimental Allergic Encephalomyelitis(EAE) is a model of multiple sclerosis and induced by administration spinal cord homogenate with complete Freund¢¥s adjuvant(inactive mycobacteria tuberculosis, Difco, co.) Glycerylphophorylcholine(GPC) phosphocholine phosphodiesterase activity reduced in EAE-induced mice and evaluated by dexamethasone, known as drug control for screening on EAE. This enzyme activity is suggested for new pharmacological screening method in EAE model. CV3988, platelet activating factor antagonist(20mg/kg) protected significantly reduction of enzyme activity. Pretreated- and posttreated- cyclosporine A decreased significantly reduction of GPC phosphocholine phosphodiesterase activity. Thymopentin 1, 5, 10mg/kg developed reduction of enzyme activity.
